This Leica TCS SP5 System covers a wide variety of confocal applications delivering bright, noise-free images with minimal photo damage at high speed. It provides the full range of scan speeds at the highest resolution with a high-efficiency five channel SP photomultiplier for simultaneous detection of different fluorescent dyes, plus a tunable filter for laser line selection and intensity control with microsecond precision.
The system uses an upright Leica DMI 6000 B microscope with both a standard stage and a galvanometric scanning stage with a 21.2mm (SFZ) Diagonal large scanning field and variable speeds from up to 2800 Hz in bidirectional mode. Connected to the microscope are multiple laser lines for the excitation of fluorophores: Multiline Argon Laser (458nm, 476nm, 488nm, 496nm, 514nm), HeNe Laser (543nm), and HeNe Laser (633nm); plus there are two photomultiplier Spectral Detector SP fluorescence/reflection detectors for simultaneous detection of different fluorescent dyes, one transmitted light detector for acquisition of DIC or bright field images, two motorized sliders for transmitting a step-lessly tunable band to the sensor, one imaging laser RGB for dichroic selection, and one magnification changer booster lens for the fluorescence axis. Also included are five objective lenses for fluorescence and confocal imaging at a range of 10X to 100X magnification: HC PL Fluotar 10x/0.30 dry, HC PL APO CS 20x/0.70 dry, HCX PL Fluotar L 40x/0.60 Corr 0-2/C dry, HCX PL APO L U-V-I 63x/1.20 water immersion, and HCX PL APO CS 100x/1.4-0.7 oil immersion. To further improve work flow autoselected inline filter cubes are built into the microscope are for safe direct viewing of laser excited samples in the UV (340-380nm), blue (450-490nm), green (515-560nm), and red (analyzer) range. The high imaging capability is also maximized by attachment of the microscope to a passive vibration isolation table designed to cancel environmental vibrations and prevent noisy images.
High speed live data collection is accomplished at a HP xw8400 workstation using Leica’s Application Suite Advanced Fluorescence (LAS AF) software version 1.8.2 build 1465 with 3D visualization. This suite controls all scanner and image acquisition parameters for recording manual and automatic workflows at up to 54 Images/second; then provides tools for measuring, processing, organizing and archiving data sets.
